越来越多的研究表明,发酵乳杆菌可以用于溃疡性结肠炎的预防和治疗。本研究中,我们从中国不同地区的人群粪便样本中分离出了105 株发酵乳杆菌,并对其基因组草图进行了测序。我们分析了这些菌株的泛基因组和系统发育特征,并对4 个模型菌株(发酵乳杆菌3872、CECT5716、IFO3956 和VRI003)也进行了分析。系统发育分析表明,发酵乳杆菌基因组的进化方向与宿主的地理位置、性别、族群和年龄没有明显的关系。我们挑选了3 株来自不同的系统发育支系的发酵乳杆菌(FWXBH115、FGDLZR121和FXJCJ61)和发酵乳杆菌模式菌株CECT5716,通过构建右旋糖酐硫酸钠(DSS)诱导的结肠炎小鼠模型,
探究这几株菌的抗炎和免疫调节活性。发酵乳杆菌FXJCJ61 和CECT5716 可以通过缓解所有结肠炎相关的组织学指标,保护黏膜完整性,增加肠道短链脂肪酸(SCFA),显著减轻结肠炎,而其他两株菌未能提供类似的保护作用。发酵乳杆菌FXJCJ61 和CECT5716 的抗炎机制与核转录因子kappa-B(NF-κB)信号通路激活以及促进白细胞介素10(IL-10)的产生有关。比较基因组分析结果表明,这些有益发酵乳杆菌的抗炎作用可能与一些特定基因有关。

With the abuse of antimicrobial agents in developing countries, increasing number of carbapenem-resistant Enterobacteriaceae (CRE) attracted considerable public concern. A retrospective study was conducted based on 242 CRE strains from a tertiary hospital in Hangzhou, China to investigate prevalence and drug resistance characteristics of CRE in southeast China. Bacterial species were identified. Antimicrobial susceptibility was examined by broth microdilution method or epsilometer test. Resistant β-lactamase genes were identified by polymerase chain reaction and sequencing. Genotypes were investigated by phylogenetic analysis. and were the most prevalent types of species, with occurrence in 71.9% and 21.9% of the strains, respectively. All strains exhibited high resistance (>70%) against β-lactam antibiotics, ciprofloxacin, trimethoprim–sulfamethoxazole, and nitrofurantoin but exhibited low resistance against tigecycline (0.8%) and minocycline (8.3%). A total of 123 strains harbored more than two kinds of β-lactamase genes. , , , and were the predominant genotypes, with detection rates of 60.3%, 61.6%, 43.4%, and 16.5%, respectively, and were highly identical with reference sequences in different countries, indicating potential horizontal dissemination. IMP-4 was the most frequent class B metallo-lactamases in this study. In conclusion, continuous surveillance and effective prevention should be emphasized to reduce spread of CRE.

单相鼠伤寒沙门菌(Salmonella enterica serovar 4,[5],12:i:-, S. 4,[5],12:i:-)作为鼠伤寒沙门菌的变种,已成为全球公共卫生领域备受关注的新兴血清型,其在中国的基因组流行病学特征尚不明确。本研究对2008-2017年分离自腹泻患者的100株序列型34型(ST34)S. 4,[5],12:i:-开展的流行情况、耐药表型和群体基因组学的分析发现:多数菌株对氨苄西林、链霉素、磺胺类和四环素耐药(耐药表型ASSuT);基于贝叶斯系统发育分析,中国分离株起源于欧洲谱系,进化为携带不同耐药基因和质粒的两个分支。进化支1中gyrA基因点突变和质粒介导喹诺酮耐药基因携带率较高,而进化支2中多数菌株fljBA操纵子插入了耐药编码区域[编码ASSuT耐药表型,包括blaTEM-1B(氨苄西林耐药)、strAB(链霉素耐药)、sul2(磺胺类耐药)和tet(B)(四环素耐药)],导致了血清型单相变异。IncHI2质粒是该血清型中导致细菌严重耐药的主要可移动遗传元件,普遍存在于进化支1的分离株中。综上,多重耐药S. 4,[5],12:i:-菌株在中国的流行情况日益严重,且进化为携带不同耐药基因的多个谱系,需采取有效措施控制此人畜共患病原体的传播,并基于“同一健康”策略开展监测及制定干预策略。

Wind turbine gearbox (WTG), which functions as an accelerator, ensures the performance and service life of wind turbine systems. This paper examines the distinctive modal properties of WTGs through finite element (FE) and experimental modal analyses. The study is performed in two parts. First, a whole system model is developed to investigate the first 10 modal frequencies and mode shapes of WTG using flexible multi-body modeling techniques. Given the complex structure and operating conditions of WTG, this study applies spring elements to the model and quantifies how the bearings and gear pair interactions affect the dynamic characteristics of WTGs. Second, the FE modal results are validated through experimental modal analyses of a 1.5 WM WTG using the frequency response function method of single point excitation and multi-point response. The natural frequencies from the FE and experimental modal analyses show favorable agreement and reveal that the characteristic frequency of the studied gearbox avoids its eigen-frequency very well.
